TIMOTHY P. MORAN: Characterization of Dendritic Cells Transduced with Venezuelan Equine Encephalitis Virus Replicon Particles as Therapeutic Cancer Vaccines (Under the direction of Jonathan S. Serody and Robert E. Johnston) Cancer vaccines seek to harness the specificity of T and B lymphocytes for reduction of tumor burden, as well as prevention of recurrent disease by establishing immunological memory. Because of their ability to initiate adaptive immune responses, dendritic cells (DCs) presenting tumor antigens have frequently been used as cancer vaccines. Unfortunately, induction of therapeutic responses in cancer patients has been sporadic, suggesting that current DC vaccines are unable to surmount tolerance against tumor antigens. The transduction of DCs with recombinant viral vectors may be a viable strategy for augmenting the ability of DC vaccines to break tolerance, as this approach can be used to efficiently deliver tumor antigens to DCs in the context of an immunostimulatory viral infection. Therefore, we have investigated the potential of DCs transduced with Venezuelan equine encephalitis virus replicon particles (VRPs) as cancer vaccines. VRPs could efficiently transduce human and murine immature DCs ex vivo, leading to high-level transgene expression, DC maturation, secretion of proinflammatory cytokines and efficient presentation of VRP-encoded antigens to T cells. VRP-transduced DCs (VRP-DCs) expressing a truncated neu oncoprotein stimulated neu-specific T cell and antibody responses and induced regression of established tumors in nontolerant mice. In contrast, VRP-DCs failed to induce robust antitumor responses in mice tolerant to neu, and were likewise unable

Alphavirus vector-based replicon particles expressing multivalent cross-protective Lassa virus glycoproteins

Lassa virus (LASV) is the most prevalent rodent-borne arenavirus circulated in West Africa. With population at risk from Senegal to Nigeria, LASV causes Lassa fever and is responsible for thousands of deaths annually. High genetic diversity of LASV is one of the challenges for vaccine R&D. We developed multivalent virus-like particle vectors (VLPVs) derived from the human Venezuelan equine ence...

Eradication of established tumors by vaccination with Venezuelan equine encephalitis virus replicon particles delivering human papillomavirus 16 E7 RNA.

The etiological role of human papillomaviruses (HPV) in cervical and other cancers suggests that therapeutic vaccines directed against requisite viral antigens may eradicate tumors or their precursors. A Venezuelan equine encephalitis (VEE) alphavirus vector delivering the HPV16 E7 RNA was evaluated for antitumor efficacy using a murine E7+ tumor model. Vaccination with VEE replicon particles e...

Systemic, mucosal, and heterotypic immune induction in mice inoculated with Venezuelan equine encephalitis replicons expressing Norwalk virus-like particles.

Norwalk-like viruses (NLVs) are a diverse group of single-stranded, nonenveloped, positive-polarity RNA viruses and are the leading cause of epidemic acute gastroenteritis in the United States. In this study, the major capsid gene of Norwalk virus, the prototype NLV, has been cloned and expressed in mammalian cells using a Venezuelan equine encephalitis (VEE) replicon expression system. Upon in...

Multiagent vaccines vectored by Venezuelan equine encephalitis virus replicon elicits immune responses to Marburg virus and protection against anthrax and botulinum neurotoxin in mice.

The development of multiagent vaccines offers the advantage of eliciting protection against multiple diseases with minimal inoculations over a shorter time span. We report here the results of using formulations of individual Venezuelan equine encephalitis (VEE) virus replicon-vectored vaccines against a bacterial disease, anthrax; a viral disease, Marburg fever; and against a toxin-mediated dis...

Ebolavirus Vaccines: Progress in the Fight Against Ebola Virus Disease.

Ebolaviruses are highly infectious pathogens that cause lethal Ebola virus disease (EVD) in humans and non-human primates (NHPs). Due to their high pathogenicity and transmissibility, as well as the potential to be misused as a bioterrorism agent, ebolaviruses would threaten the health of global populations if not controlled. In this review, we describe the origin and structure of ebolaviruses ...
